Drug transporters are expressed in many tissues such as the intestine, liver, kidney, and brain, and play key roles in drug absorption, distribution, and excretion. MDCKII cells are the classical model to study active transport of compounds. Besides passive cell permeability, active transport is the major process affecting drug delivery.
A transport substrate could either increase or decrease drug delivery to the target tissue. For this reason, study of the affinity of a given compound for the most known uptake and efflux transporters should be addressed during Drug Discovery. MDCKII cells overexpressing different transporters are also classically used to assess potential drug interactions. During the ADME studies, definition of potential interactions between drugs is a key factor to determine possible problems in drug delivery or cytotoxic effects, and therefore these studies are demanded and regulated from agencies like FDA and EMA.
